چکیده

Department of Biological Sciences, University of Hull, Hull HU6 7RX, UK Malic enzyme was shown to be a major source of NADPH for the synthesis of storage I ipid by Aspergihs nidulsns. A previously produced mutant lacking malic enzyme activity (scuK248) accumulated only half the lipid (l2%, w/w, of cell dry weight) accumulated by strains of A. nidulans possessing malic enzyme. When cultivated under conditions designed not to promote the production of storage lipid all three strains of A. nidulans studied contained equivalent amounts of lipid (5%, w/w@ of cell dry weight). Thus malic enzyme did not appear to be limiting the synthesis of metabolically active lipid (e.g. membrane lipids). No evidence of a role for malic enzyme in the desaturation of fatty acids was found.

Kinetic studies of the induction of nitrate reductase and cytochrome c reductase in the fungus Aspergillus nidulans.

In an earlier paper (Cove, 1966) it was reported that the kinetics of appearance of nitrate reductase (NADPH-nitrate oxidoreductase, EC 1.6.6.3) on the addition of nitrate to a growing culture of Aspergillus nidulans were different in certain respects from those found for many Escherichia coli enzymes. When urea is used as an initial nitrogen source, a further difference is found: enzyme synthe...

The pivotal role of malic enzyme in enhancing oil accumulation in green microalga Chlorella pyrenoidosa

BACKGROUND The fast growing photosynthetic microalgae have been widely used in aquaculture, food, health, and biofuels. Recent findings in the diatom has proposed a pivotal role of NADP-malic enzyme in generation of NADPH as an important supply of reducing power for fatty acid biosynthesis. To test the lipogenic malic enzyme for fatty acid synthesis in green algae, here the malic enzyme gene Pt...

Characterization of the reduced nicotinamide adenine dinucleotide phosphate-nitrate reductase of Aspergillus nidulans.

The reduced nicotinamide adenine dinucleotide phosphate (NADPH)-nitrate oxidoreductase (EC 1.6.6.2) from Aspergillus nidulans was purified over 200-fold by use of salt fractionation, gel filtration, and ion-exchange chromatography. The purified enzyme was specific for NADPH and catalyzed reduction of nitrate, cytochrome c from isolated mitochondria of Aspergillus, and mammalian cytochrome c. An...

Regulation of lipid accumulation in oleaginous micro-organisms.

A small number of eukaryotic micro-organisms, the oleaginous species, can accumulate triacylglycerols as cellular storage lipids, sometimes up to 70% of the biomass. Some of these lipids, particularly those containing high proportions of polyunsaturated fatty acids of nutritional and dietary importance, are now in commercial production; these are known as single-cell oils. The biochemistry of l...

Co-Cultivation of Penicillium sp. AKB-24 and Aspergillus nidulans AKB-25 as a Cost-Effective Method to Produce Cellulases for the Hydrolysis of Pearl Millet Stover

Hydrolysis of cellulose and hemicelluloses into fermentable sugars is the primary step for the production of fuels and chemicals from lignocellulosic biomass, and is often hindered by the high cost of cellulolytic and hemicellulolytic enzymes. In the present study coand monocultures of Penicillium sp. AKB-24 and Aspergillus nidulans AKB-25 were used under a variety of fermentation conditions to...
